TPX2和胸苷磷酸化酶在胃癌中的表达及其临床意义

摘要

目的:探讨Xk1p2靶蛋白(targeting protein for Xk1p2,TPX2)和胸苷磷酸化酶(thymidine phosphorylase,TP)在胃腺癌中的表达及其与临床病理因素的关系。方法:应用免疫组织化学SP法,检测75例胃腺癌(胃腺癌组)、33例异型增生(癌旁异型增生组)和75例正常胃黏膜组织(正常胃黏膜组)中TPX2和TP蛋白的表达。结果:胃腺癌组TPX2和TP的阳性表达率分别为70.7% 和62.7%;癌旁异型增生组为33.3%和39.4%;正常胃黏膜组为2.7% 和5.3%。TPX2和TP在正常胃黏膜组、癌旁异型增生组及胃腺癌组中的表达率依次增高,经多个样本率的比较,3组间均有统计学意义(P<0. 01)。TPX2蛋白的表达与肿瘤的分级、分期、浸润深度及淋巴结转移有关(P<0.05),与年龄、肿瘤大小无关(P>0.05)。TP的阳性表达率与肿瘤的分期、浸润深度、淋巴结转移及肿瘤大小均有统计学意义(P<0.05),与年龄、分化程度无关(P>0.05)。TPX2与TP在胃腺癌中的表达呈正相关关系(r=0.4697,P<0.01)。结论:TPX2和TP可能共同参与了胃腺癌的发生、发展及转移。联合检测TPX2和TP有望成为胃腺癌的早期诊断及预后判断的候选标志物。

Expression and clinical significance of targeting protein for Xk1p2 and thymidine phosphorylase in gastric carcinoma

Abstract:

Objective To study the expressions of targeting protein for Xk1p2 (TPX2) and thymidine phosphorylase(TP) in gastric carcinoma and to evaluate its correlation with the clinicopathological features. Methods Immunohistochemical staining was used to detect the expressions of TPX2 and TP in tissues from 75 cases of gastric carcinoma (gastric carcinoma group),33 cases of atypical hyperplasia(atypical hyperplasia group) and 75 cases of normal stomach mucosa (normal stomach mucosa group). Results The positive expression rates for TPX2 and TP protein in normal control, atypical hyperplasia and gastric groups were 2.7% and 5.3 %, 33.3% and 39.4 %, 70.7% and 62.7 %, respectively. There was a significant statistic difference among the 3 groups (P<0.05). The positive rate of TPX2 was closely related to the depth of invasion, lymph node metastasis,histological grade and tumor stage (P<0.05) except for age and tumor size whereas the positive rate of TP was closely related to depth of invasion, lymph node metastasis, and tumor stage, tumor size (P<0.05) except for age and histological grade. There was positive correlation between TPX2 and TP in gastric carcinoma (r=0.4697, P<0.01). Conclusion High expression of TPX2 protein together with TP protein might play important roles in the tumorigenesis, progression and metastasis of gastric carcinoma. Combined examination of TPX2 and TP might be useful in the early diagnosis and for the prognostic judgement of gastric carcinoma.
